The antigen carrier is a synthetic peptide construct (E-coil) containing the modified amino acid norleucine (X). The peptide antigen forms a loop via an internal disulfide bridge, and is covalently bound at the amino terminus to the carrier by specific labeling of a free cysteine in the carrier with iodoacetylated peptide. Antiserum from a second rabbit did not respond to this heterotypic peptide. One of two rabbits immunized with the tetanus toxoid-conjugated peptide also responded to this heterotypic peptide with an eight-fold lower ka. .
