A panel of conformation-dependent mAbs was tested by radioimmunoprecipitation. The neutralizing mAb H2.1 lost binding reactivity to HeV G glycoprotein mutants with alanine substitution at epitope residues. The epitope (and additional D257, D260, G439, G449, and D468) residues are critical for HeV G binding to its receptors ephrinB2 and ephrinB3, and interaction with HeV F for membrane fusion.
