Residue Asp63, in site E, was shown to be critical for the binding of mAb HC45 to the viral hemagglutinin by selection of a viral variant with an Asp63 to Asn substitution. Variant virus HA protein did not block the mAb in a hemagglutination inhibition assay. Binding of mAb HC45 to the virus was independent of the pH-dependent conformation of HA.
