The binding of mAbs to the AD-1 domain of HCMV gB (552-635) was analyzed using a panel of bacterial fusion proteins expressing sequences between 549 and 635 of gB harboring point mutations. Mutations involving the cysteine residues at positions 573 and 610, as well as proline residues at positions 570, 577 and 613 resulted in loss of binding. .
