AR2 scFv was obtained by selecting yeast-displayed mutants of the humanized C25 mAb by increasing the affinity for BoNT/A1. AR2 binds BoNT/A1 with very high affinity and exhibits the highest level of in vivo neutralization, but has minimal neutralizing activity for BoNT/A2. The residues involved in this functional epitope were identified by mutational analysis of the energetic contribution of the individual BoNT/A2 residues. The IgG ka to BoNT/A1 is 3.7x106 M-1 s-1 and to BoNT/A2 is 8.31x105 M-1 s-1.
