The epitope residues were derived by sequencing the N-terminal region of 35K fragments of proteins isolated from schizont and merozoite lysates.
Sera from monkeys immunized with the chimeric SPf66 vaccine reacted with solubilized P. falciparum schizont antigens in western blot analysis. The reactivity with the cleavage products shown on the gel were consistent with the fragments from which the epitope was derived. The use of QS-21 as adjuvant with SPf66 instead of Freund's notably increased antibody titers.
