The intact 9-40 antibody was derived from the same idiotypic family as 4-4-20 (see complex of 4-4-20 Fab with fluorescein PDB ID 4fab) but was altered by fewer somatic mutations in the germline genes which led to a 1000-fold reduction in affinity for fluorescein by 9-40 relative to 4-4-20.
