This epitope is a nonamer consisting of D-glutamic acid residues linked at the gamma-COOH groups. It is derived from the capsule of Bacillus anthracis, which is a polymer of gamma-D-glutamic acid.
The assay was a flow cytometric opsonophagocytosis assay. RAW 264.7 cells that had taken-up bacilli gave a fluorescent peak two orders of magnitude brighter than background fluorescence. Levels of phagocytosis were expressed as percentage of fluorescence-positive gated RAW 264.7 cells. The level of opsonophagocytosis was reduced by inhibition with both the epitope, and with B. licheniformis poly-glutamic acid. Specificity of antibodies was evaluated using a serum pool prepared by combining serum samples from mice that had received two injections. Fluorescein-labeled heat-killed encapsulated B.anthracis bacilli (Ames strain) were incubated with various dilutions of an antibody followed by addition of RAW 264.7 cells.
