The minimal conformational epitope was not fully identified with certainty. However, based on experimental findings, it was shown that the HPV16 L1 loop domain I and possibly domain II are involved in the binding of monoclonal antibody 16A. Conformational epitope. The position relative to the protein in genbank is 40-52, 113-140.
Binding of a monoclonal antibody to the conformational epitope of HPV16.L1 protein was demonstrated by the expression of wild type and chimeric recombinant L1 in SF9 cells infected by recombinant bacculoviruses. The region targeted by the 16A monoclonal antibody was identified as fragments I and II by exchanging protein fragments in 2 different constructs.
