Neutralizing peptide ligands selected from phage-displayed libraries mimic the conformational epitope on domain III of the Japanese encephalitis virus envelope protein.
|
| description: |
This peptide is shown to mimic envelope (E) protein from Japanese encephalitis virus (JEV) in its binding to a neutralizing antibody and by eliciting neutralizing antibodies. 12-mer peptide Phage display library was screened with mAb E3.3 with 3 rounds of biopanning to find peptides able to induce a protective response against JEV.
A slight, but above background reactivity of mAb E3.3 to this phage clone was observed. A similar result was seen in a competitive ELISA for inhibition of binding of the mAb to E protein. A phage particle expressing the epitope also inhibits neutralization of JEV by mAb E3.3.
