The assay investigated the ability of the monoclonal antibody to neutralize the hemolytic activity of listeriolysin by incubation with listeriolysin, adding sheep erythrocytes, and measuring the amount of hemoglobin released. The neutralization activity was reported as strong. To distinguish whether the neutralizing antibody inhibited binding of listeriolysin to the cellular membrane or a subsequent step, the monoclonal antibody was preincubated with listeriolysin at neutralizing concentrations and this complex was incubated with the mouse fibrblast cell line, 36.2.1. It was shown that while hemolytic activity was inhibited, the monoclonal antibody was able to bind to the fibroblasts via listeriolysin, suggesting that neutralization was not mediated by binding of listeriolysin to the cellular membrane.
