This linear determinant is deduced from reactivity of overlapping 12-mers in serum of 1 of 59 patients positive for DFS70 antibodies. It was determined that the primary autoantibody determinant is alpha-helical, conformation-dependent, and restricted to amino acids 349-435 of DFS70, but specific residues comprising the epitopic determinant(s) were not refined in this publication.
Epitope mapping of linear determinants was performed via peptide array of 12-mer peptides spanning DFS70 residues 326-527. In this experiment, the array of peptides was tested against 59 samples of human sera. Only 1 of the 59 sera tested were reactive against the array. In the one reactive sample, there were a total of 5 reactive spots, all corresponding to this C-terminal epitope.
