S residues were substituted for the natural C residues at positions 16 and 18 to prevent disulfide bonding. The paper uses the A/PR/8/34/Mount Sinai strain, but the sequence of the M2e peptide is not found in this strain. The Swiss Prot ID given is for Influenza A virus (A/USSR/90/77(H1N1). Several flu strains contain this sequence.
The authors specify that mAb 14C2 is specific to this epitope. Multi-epitope constructs consisting of 1 copy of M2e (a matrix protein 2 ectodomain epitope from influenza A virus), 1 copy each of S1 and S2 (hemagglutinin peptides) were used as antigens.
