The Cysteine residues present at positions 4 and 8 in the native sequence were substituted by aminobutiric acid, which has the same hydrophobic, steric, and electronic properties as Cysteine, but avoids the secondary reactivity of thiol groups during the conjugation to liposomes.
The strongest interactions were between antibody with free peptide, and with peptide covalently coupled to liposome by a thioether bond. Disulfide and amide containing bonds yielded lower responses. The ligand used was an IgG anti-epitope purified by affinity chromatography.
