This epitope was identified by screening of synthetic peptide libraries, and was found to mimick an antigenic determinant of hepatitis A virus. No sequence homology was found between the epitope and the hepatitis A virus capsid proteins VP1 and VP3.
Synthetic hexapeptide libraries with the structure XXXXXXGGG-PAM resin were synthesized using processes of division, coupling, and recombination of individual resins. The library was screened with anti-HAV monoclonal antibodies in order to determine their epitope specificities. This process led to the identification of a peptide (DVMYLKGGG), which was later optimized to MWLDVMYLKGGG by a secondary semirandom library. Both peptides were synthesized in tetrameric form and were shown to react with the monoclonal antibodies. In this assay, tetrameric peptides were shown to react with human polyclonal anti-HAV antibodies.
