To determine the location of epitopes on the hemaglutinnin molecule of the seal influenza virus, antigenic variants selected in the presence of monoclonal antibodies were analyzed. All variants had single amino acid changes. In a subsequent assay, the monoclonal antibody defining this epitope inhibited the fusion of the virus with the intracellular vacuolar membrane of MDCK cells, thus indicating that it did not inhibit attachment or internalization of the virus, but did inhibit the low pH-induced conformational change in the hemaglutinnin molecule, a prerequisite for the fusion step in the penetration process.
