Analysis of antibody-antigen and antibody-anti-(idiotypic antibody) cross-reactivity using synthetic peptide probes.

| description: |
This epitope is recognized by MAb 1/1. It is thought to be mimicked by an anti-idiotypic antibody (MAb 4C1) raised against the MAb 1/1. The FR2 region of MAb 4C1 harbor non-consecutive residues that may provide a sequence spatially equivalent to NXXEKQS (where S is a conservative substitution of T). Actual mimicry of the epitope is probable since MAb 4C1 is able to inhibit the interaction between MAb 1/1 and the epitope. Sequence of the X-31 virus hemagglutinin (HA) used in this study corresponds to the one reported in the pdb entry. The Swiss-prot database entry for the HA precursor from the X-31strain doesn't harbor this sequence, but the Swiss-prot entry P03437 for the parental strain Aichi/2/68 (from which the external proteins of the reassortant strain x-31 are derived) does.
Affinity for the immunizing peptide of MAb 1/1 (previously shown to recognize this epitope), was determined in ELISA, with two methods of data analysis yielding KD values of ~300 nM.
