This key residues from antigenic site 3 were identified by sequence analysis of viral escape mutants. The amino acid positions provided in the manuscript do not correspond to the sequence provided in the SwissProt entry cited.
Viral escape mutants of the parent virus were selected for their resistance to neutralization by the monoclonal antibody, and their hemagglutinin-esterase glycoprotein genes were subsequently sequenced. Four distinct variants were selected, however, one variant retained the capacity to bind the selecting monoclonal antibody despite its resistance to neutralization. Amino acid changes at residues 178 and 212 were also shown to decrease hemagglutinating activity.
