| name: |
Production of murine monoclonal antibodies against sulcofuron and flucofuron by in vitro immunisation.
|
| description: |
This hapten name stands for 3,4-dichloroaniline. It is used as a surrogate of the pesticide Sulcofuron (5-chloro-2-[4-chloro-2-[[[(3,4-dichlorophenyl)amino]carbonyl]amino]phenoxy]benzenesulfonic acid) for antibody screening.
Culture supernatants of polyclonal hybridomas, or of a monoclonal IgM, generated from Sulcofuron-in vitro-immunized splenocytes, are shown to recognize the related molecule DCA (3,4-dichloroaniline) bound to ELISA plates through a BSA carrier, but not other haptens-BSA or BSA-alone sensitized plates. This result shows the success of the in vitro immunization procedure, and confirms the cross-reactivity between Sulcofuron and DCA. DCA was used for antibody screening since Sulcofuron structure was unsuitable for covalent bonding to carrier proteins.
