In the paper, amino acid residues are numbered according to the alignment of different IBV S1 proteins presented in Kusters et al. (1989) Virology 169:217 [PMID: 2466369]. The alignment introduces spaces in the sequences compared. The actual position of the amino acid(s) as indicated in the paper therefore differs from the D207 IBV S1 sequence in the database.
A sandwich ELISA was used in which IBV virions in egg allantoic fluid were bound to mAb adsorbed to plates. Strain D207 IBV bound to mAb, but an escape mutant identified in neutralization assays using the same mAb did not. Sequencing of the escape mutant identified one amino acid change from Gly 291 to Glu. Unrelated mAbs still bound to the escape mutant in the assay, suggesting that the alteration did not dramatically alter protein conformation and could be involved in recognition.
