The major antigenic site 1 of the poliovirus type 1 VP1 protein was previously identified.
A chimeric poliovirus was constructed using type 1 virus with the major antigenic site 1 of type 3 VP1 protein replacing that of the type 1 VP1 site. Reactivity of the chimeric virus was tested against type-specific monoclonal antibodies in an antigen-blocking assay. One type 1-specific mAbs were capable of reacting with the type 1 virus and not the chimeric virus containing the inserted type 3 epitope. This suggested that the type 1 site was lost in chimeric virus and that the mAb 955 was specific to this peptide epitope. These mAbs gave parallel results in neutralization tests performed separately.
