Hepatitis C virus (HCV)-specific antibody producing B lymphocytes were induced in vitro with a HCV-derived lipopeptide containing a T helper cell epitope. Purified B cells and autologous T helper cells from healthy donors were cultured in vitro with the lipopeptide, and the number of IgG, IgA, and IgM antibody-producing B cells after in vitro immunization was quantified by ELISPOT. The lipopeptide was formed by coupling the peptide to a lipid tail consisting of three palmitoylacid chains. A response was also found to the lipidated peptide in 1 of 14 donors.
