Identification and assessment of new vaccine candidates for group A streptococcal infections.

Opsonization activity towards bacteria from reference strain of Streptococcus (M1GAS) and of the clinical isolate 88-30 is detected in sera of immunized outbred mice. Nevertheless, sera from mice immunized with carrier (KLH) alone also show opsonization activity (and bind the bacterial cells as detected by immunofluorescence). The reactivity in KLH alone-immunized mice sera is not significantly different from that in sera from mice immunized with peptide-KLH, but is consistently lower. The authors thus conclude that some enhancement of the activity can be attributed to the presence of peptide, but that on the other hand KLH may be useful as a carrier for anti-GAS vaccines since it has cross-reactive epitopes with GAS. Non-specific IgG binding to GAS Fc receptors is discarded as the cause for the cross-reactivity by preincubation with human IgG, after which the reactivity remains.
