The peptide was selected as one of six subregions containing substitutions (at residues 202 and 204) between mouse and hamster sequences. The peptide sequence is identical in humans.
The antibody reacts with mouse PrPsc but not hamster PrPsc by ELISA and Western blotting. The authors conclude that Mo-V is a species specific epitope. The Obihiro strain of mouse-passaged scrapie agent and the Sc237 strain of hamster-passaged scrapie agent were intracerebrally inoculated into SLC/ICR mice and Std/Syrian hamsters. The PrP enriched fractions were prepared from brains by sarcosyl extraction, differential extraction, and proteinase K treatment.
