Selection of antigenic and immunogenic mimics of hepatitis C virus using sera from patients.

This peptide was selected from a phage 9-mer library for its specific reactivity with HCV patients sera. It was shown to mimic antigenically the NS4b protein 1904-1959 fragment, and homology with its sequence can be found for residues 1931-1938 (PTHYVPESD). The positions are based on the polyprotein as found in Swiss-prot ID P26663. This peptide is derived from a phage library where the peptide sequence flanked by a cysteine residue at both ends is inserted in the pVIII major coat protein of phage particle. The cysteine residues promote intrachain disulfide bonding that constrains to some extent the conformation of the displayed peptide.
The phage carrying this peptide was initially selected as one of the a binder of a HCV patient sera Abs directly bound to the surface of polystyrene beads, followed by plaque detection by another patient's sera. The specificity of the reaction was confirmed by ELISA with a large panel of patients and normal sera. This phage was recognized by 11/44 HCV patients sera IgGs, while none of the ~50 normal sera samples had reactivity against it. It was later shown to mimic the NS4b protein region 1904-1959 antigenically, since a patient's Abs purified by immunoabsortion to this phage was able to bind the GST-NS4b(1904-1959) fusion protein.
