This peptide was selected from a phage 9-mer library for its specific reactivity with HCV patients sera. It was shown to mimic antigenically and immunogenically the NS4b protein 1904-1959 fragment, and homology with its sequence can be found for residues 1931-1938 (PTHYVPESD). The positions are based on the polyprotein as found in Swiss-prot ID P26663. This peptide is derived from a phage library where the peptide sequence flanked by a cysteine residue at both ends is inserted in the pVIII major coat protein of phage particle. The cysteine residues promote intrachain disulfide bonding that constrains to some extent the conformation of the displayed peptide.
Significant reactivity against GST-NS4b(1904-1959) is seen in mice (Balb/C, MF1 and C57BL6) immunized with the phage expressing this mimotope. Also, significant inhibition of that reactivity is seen by both the phage-peptide and by the same fusion protein in soluble competition studies, implying the reactivity obtained is mediated by peptide reactive antibodies.
