Selection of antigenic and immunogenic mimics of hepatitis C virus using sera from patients.

This peptide was selected from a phage 9-mer library for its specific reactivity with HCV patients sera. It was shown to mimic antigenically and immunogenically the HCV core protein 1-124 fragment corresponding to the core protein. Homology to its sequence can be found for residues 9-17 (KKTKRNTNR). The positions are based on the polyprotein as found in Swiss-prot ID P26663. This peptide is derived from a phage library where the peptide sequence is inserted in the pVIII major coat protein of phage particle.
Significant reactivity against GST-core(1-124) is seen in mice (Balb/C, MF1 and C57BL6) immunized with the phage expressing this mimotope. Also, significant inhibition of that reactivity is seen by both the phage-peptide and by the same fusion protein in soluble competition studies, implying the reactivity obtained is mediated by peptide reactive antibodies.
