Selection of antigenic and immunogenic mimics of hepatitis C virus using sera from patients.

This peptide was selected from a phage 9-mer library for its specific reactivity with HCV patients sera. It was shown to mimic antigenically and immunogenically the HCV core protein 1-124 fragment corresponding to the core protein. Homology to its sequence can be found for residues 10-18 (KTKRNTNRR). The positions are based on the polyprotein as found in Swiss-prot ID P26663. This peptide is derived from a phage library where the peptide sequence is inserted in the pVIII major coat protein of phage particle.
The phage carrying this peptide was initially selected as one of the a binder of a HCV patient sera Abs directly bound to the surface of polystyrene beads, followed by plaque detection by another patient's sera. The specificity of the reaction was confirmed by ELISA with a large panel of patients and normal sera. This phage was recognized by 32/43 HCV patients sera IgGs, while none of the ~50 normal sera samples had reactivity against it. It was later shown to mimic the NS4b protein region 1-124 antigenically, since a patient's Abs purified by immunoabsortion to this phage was able to bind the GST-NS4b(1-124) fusion protein.
