This peptide was previously found to be one of the 4 PrP peptides that stimulated T cell proliferation in Prnp-/- mice immunized with plasmid DNA encoding mouse PrP.
Low titer Ab responses were obtained in mice immunized with this peptide along with CpG and IFA against the immunizing peptide in ELISA. Immunization with the peptide in CFA did not produce responses in 3 animals. Isotype analysis of the response in 4 animals showed that IgG2b was found in all the animals, while IgG1 and IgG2a were found only in 0 and 1 animals respectively. Analysis of the frequency of IgG-producing B-cells in spleen and lymph node of immunized animals by limiting dilution analysis showed that the overall frequency was low, with a maximum of 1/20000 in the lymph node of one animal.
