Serial deletions of myosin-II C-terminal tail were tested for monoclonal antibody binding. Monoclonal antibodies M2.9 (isotype IgG1) bound to the epitope with inhibition of actin-activated Mg2+-ATPase and contraction of gelled cytoplasmic extracts. M2.9 poorly inhibited polymerization. Electron microscopic localization of antibody binding sites correlated well with deletion mapping. M2.9 and M2.31 compete each other for binding to myosin-II in a solid phase assay.
