Serial deletions of myosin-II C-terminal tail were tested for monoclonal antibody binding. Monoclonal antibodies M2.14 (isotype IgG1) and M2.15 (IgG1) bound to the same epitope. M2.15 inhibits contraction of gelled cytoplasmic extracts, but has no effect on actin-activated Mg2+-ATPase and polymerization. Electron microscopic localization of antibody binding sites correlated well with deletion mapping. M2.14 and M2.15 compete each other for binding to myosin-II in a solid phase assay.
