Serial deletions of myosin-II C-terminal tail were tested for monoclonal antibody binding. Monoclonal antibody M2.36 (isotype IgG2b) bound to the epitope with no effect on actin-activated Mg2+-ATPase, and inhibition of contraction of gelled cytoplasmic extracts. M2.36 partially inhibited the formation of dimers. Electron microscopic localization of antibody binding sites correlated well with deletion mapping. M2.36, M2.39 and M2.50 compete each other for binding to myosin-II in a solid phase assay.
