Serial deletions of myosin-II C-terminal tail were tested for monoclonal antibody binding. Monoclonal antibody M2.38 (isotype IgG1) bound to the epitope with no effect on actin-activated Mg2+-ATPase, and weak inhibition of contraction of gelled cytoplasmic extracts. Electron microscopic localization of antibody binding sites correlated well with deletion mapping.
