The epitope was identified based on nucleotide homology to a HEV-specific antigen-producing clone from HEV strain Mexico. A clone encoding the epitope was generated by PCR amplification of an HEV strain Burma cDNA and cloned into a lambda gt11 vector. Immunoscreening by Western blotting or in a plaque assay format showed that the expressed epitope was recognized by 4 acute- and 2 convalescent-phase human sera collected during HEV outbreaks in Burma, Somalia, and Mexico. The authors suggest that the epitope is broadly reactive and represents a HEV type-common epitope.
