Affinity for the whole toxin was determined for a purified sc-Fv fragment selected from a phage display library constructed with the VH and VL cDNAs from a mice immunized with the Hc (receptor-binding) domain of BoNT/A followed by a boost with the holotoxin. This scFv was shown to bind Hc domain in ELISA and to have neutralizing activity of the holotoxin in an in vitro mouse hemidiafragm assay.
