Affinity for the Hc domain was determined for a purified scFv fragment selected from a phage display library derived from a pentavalent toxoid-immunized donor. This scFv is also shown to bind Hc domain in ELISA and to have neutralizing activity of the holotoxin in an in vitro mouse hemidiafragm assay. 2 other scFv were found to be clonally related to this one by sequence analysis and to bind to the same or overlapping epitopes (referred to as cluster I) by competition for reactivity on the SPR chip.
