Epitope mapping was performed using a library of 100 overlapping dodecamers, shifted by 2 amino acids, spanning residues 23-232 of the mature mouse prion protein. The epitope of MAb POM4, POM10, and POM19 was deduced to include residues 121-134 and 218-221 based on the ability of dodecamers to inhibit binding of the antibody to mouse PrP(23-230).
