This fragment is expressed in vitro from a synthetic gene constructed to encode the residues 848-1278 (binding domain) of the BoNT/F toxin of strain NCTC 10281. The nucleotide sequence of the gene was modified to include an initiation methionine and to have optimal codon usage in E.coli.
Even in the absence of a measurable response by ELISA towards whole type F toxin, mice immunized 2 or 3 times with different doses (1.9 to 1000 ng) of the Hc fragment show dose and number of immunization dependance in the protection to subsequent (14 days after the last immunization) challenge with the toxin. As little as 3 doses of 3.9ng or 2 doses of 31.2ng were sufficient to produce a 100% survival rate.
