MAb 3F4, which binds to the epitope, immunoprecipitated Prpc, with or without prior denaturation in SDS, from brain lysates of uninoculated transgenic mice expressing WT mouse PrP tagged with the MAb 3F4 epitope. MAb 3F4 reacted poorly with PrPsc from brain lysates of Tg(PG14) transgenic mice expressing mutant, epitope-tagged mouse PrP, which causes a spontaneous fatal neurodegenerative illness, or Tg(PG14) mice injected with the RML isolate of scrapie. After denaturation in SDS, however, the MAb efficiently immunoprecipitated PrPsc from both transgenic mouse groups. This confirms that the epitope becomes inaccessible during the conversion of PrPc to PrPsc.
