Fab D18 recognizes the epitope which includes alpha-helix A (residues 144-153). Fab D18 binds to SHaPrPc expressed on the surface of transfected CHO cells and does not block binding of Fabs to epitopes in the N-terminal part of PrP, indicating a lack of interaction of the N- and C-terminal domains on the cell surface. Fabs D18 and 28d [to an overlapping epitope (residues 144-156)] reciprocally block binding. D18 blocks 70% binding of Fab 13A5 to an overlapping epitope (residues 138-141), but reciprocally, 13A5 only blocks D18 binding 20% because D18 binds to a larger fraction of cell surface PrP than 13A5.
