The epitope was one of 40 homogeneously distributed sequences selected by principle component analysis from a set of 234 natural HCV hypervariable region 1 matching residues 384-411 of BK strain.
Antibodies from a pool of 8 HCV infected sera were affinity purified on multiple antigenic peptide (MAP) of mimotope H1 (sequence: QTHTTGGVVGHATSGLTSLFSPGPSQK). Reactivity of equal amounts of immunopurified antibodies was measured by ELISA on the epitope synthesized as MAP. Positive reactivity is that only when average values from two independent experiments were greater than 3-fold of both the background values of two unrelated peptides and the average values from 10 non-infected human sera.
