The ten monoclonal antibodies recognize PrPSc from prion-infected brains of various species. All antibodies reacted with mouse, hamster and bovine PrPSc. All antibodies with the exception of 1A7 and 3A2 reacted with sheep PrPSc. Reactivity to human PrPSc was not determined for 20A13, 1A7, 3A2, 12A5 Mabs, whereas 1A12, 17D10,17D4, 16A18, 12B1 Mabs were reactive.In another experiment, chimeric IgG monoclonal antibodies from light and heavy chain variable regions of five IgM Mabs (16A18, 20A13, 1A7, 9A4 and 12A5) were constructed in a dog IgG framework in order to exclude the possibility that PrPSc recognition is of low affinity and is dependent on the high affinity of IgM binding sites. All recombinant IgG antibodies selectively immunoprecipitated murine PrPSc.
