Fab 3F4, which recognizes the epitope, binds to SHaPrP(29-231) independent of prior incubation of the antigen at pH 2-12. Fab 3F4reacts strongly with SHaPrP(90-231) exposed to pH <4, but reactivity decreases significantly at pH 4.7-5.2 and remains reduced over the range pH 5-12. The authors attribute the change in reactivity to a conformational change in the N-terminal region of SHaPrP(90-231).
