This amino acid was found to be a key residue for monoclonal antibody reactivity to Human herpesvirus 6A strain U1102.
To determine the amino acid residue(s) responsible for the HHV-6A-specific reactivity with the monoclonal antibody, the neutralization site was mapped to amino acids 335-395 with truncated translation products of glycoprotein B of HHV-6A. Amino acid comparison between HHV-6A and 6B in this region revealed five amino acids that are HHV-6A-specific. Point mutations were introduced at those amino acids, and reactivity with mutagenized glycoprotein B suggested that the Asn residue at position 347 of HHV-6A strain U1102 was involved in recognition.
