HCV/GBV-B chimeric proteins were used to map the epitope in HCV NS5B for the epitope-specific monoclonal antibody. This approach was based on the fact that GBV-B NS5B possesses a high degree of homology to HCV NS5B but lacks reactivity with the monoclonal antibody. The monoclonal antibody reacted with HCV NS5B 231-372, but did not react with 267-372. The results suggested that the epitope is located between amino acid 231 and 267 of HCV NS5B.
