Amino acid residues of the peptide are numbered from the N-terminal glutamic acid of mature PA.
Preincubation of PA and lethal factor (LF) with 1700 µg MAb 2D3PA or 1300 µg of MAb 2D5PA ascites Ig before injection into rats delayed death by about 18 h. No survival was observed with MAbs 10D2PA or 10G4PA. MAbs 2D3PA, 2D5PA, 10D2PA, and 10G4PA preincubated with PA and LF before addition to J774A.1 cells neutralized lethal toxin cytolytic activity. These MAbs neutralized lethal toxin cytolytic activity whether they were added before or after PA bound to cells. MAbs 2D3PA, 2D5PA, and 10D2PA inhibited binding of LF to cell-bound PA by 78-88%. MAb 10G4PA, which neutralized lethal toxin cytolytic activity only at high concentration, did not inhibit binding of LF to cell-bound PA.
