Anti peptide 89-104, a monospecific polyclonal antibody raised in rabbits, recognizes PrPCof various species. This antibody reacted with sheep, goat and dog PrP, but not with cow, man, pig, cat, mink, guinea pig, hamster and mouse PrPC.It also reacts with sheep PrPScin the absence or presence of proteinase k treatment. Only hamster brain homogenates treated with proteinase k showed a weak reactivity with the antiboby. No reactivity was observed with cow or mouse PrPSc.In addition, in antibody competition assays, when antisera were preabsorbed to the peptide prior to immunoblotting, no specific binding to PrPCwas observed.
