The antigenicity of the NS5 region with anti-HCV-positive sera was assessed using a set of 199 overlapping hexapeptides spanning residues 2530-2733 of HCV subtype 1b. The epitope was deduced to be one of six regions with significant reactivity and was designated region I. The sequence of this region varies between subtypes 1a, 1b, 2a, and 2b. Fifty-three pecent (80 of 150) of patients reacted with the epitope from at least one of three subtype 1b isolates and 39% (59 of 150) reacted with the epitope from at least one of two subtype 1a isolates.
