N2a cells were infected with the mouse-adapted scrapie strain 22L. Culturing N2a/22L cells in the presence of epitope-specific MAb 6D11 abrogated PrPsc without affecting the level of PrPc, as detected by immunoblotting PK-resistant PrPsc and total untreated PrP in cell lysates.The effect of treatment with Mab 6 D11 persisted for 14 days in the absence of antibody. MAb 6D11 did not have a trophic or toxic effect on N2a/22L cells. Preincubation of N2a cells prior to inoculation or preincubation of the inoculum (brain homogenate from 22L-infected mice) with MAb 6D11 also resulted in reduced PrPsc levels.
