The epitope of the monoclonal antibody 3A4 was determined by epitope mapping using synthetic peptides derived from the source protein (histidine rich protein, from 46 isolates of Plasmodium falciparum); substitutions of Y or V for the first or last amino acid, respectively, still allows binding to mAb 3A4. The minimal epitope, determined with glycine scanning, is AHHAHHA.
